Roxithromycin (CAS: 80214-83-1) is a semi-synthetic 14-membered macrolide antibiotic, derived from erythromycin via 9-position oximation modification. It has good acid stability and oral bioavailability, inhibiting bacterial protein synthesis to exert antibacterial activity. It is widely used in the treatment of infections caused by Gram-positive bacteria, anaerobic bacteria and atypical pathogens, and is also used as a pharmaceutical raw material compliant with USP and Chinese Pharmacopoeia standards.

Synthesis Method
- Raw Materials: Erythromycin thiocyanate, hydroxylamine hydrochloride, methoxyethoxymethyl chloride (MEMC), triethylamine, sodium methoxide, DMF, methanol[8]
- Synthesis Route: Two-step reaction – oximation of erythromycin to form erythromycin oxime, then etherification with MEMC to obtain roxithromycin[5][8]
- Reaction Steps: Dissolve raw materials, conduct oximation at 35–45 °C, then etherification at -10~-5 °C, followed by precipitation, washing, drying and purification to get the product[8]
