Product Description
Sucralfate (CAS: 54182-58-0), also known as sucralfate aluminum, is a basic aluminum salt of sucrose octasulfate, belonging to the class of gastric mucosal protective agents. It appears as a white to slightly yellowish white amorphous powder, odorless, tasteless, and hygroscopic. As a locally acting medication, it reacts with hydrochloric acid in the stomach (at pH < 4) to form a cross-linked, viscous paste-like protective film, which is widely used in the pharmaceutical industry for the treatment of gastrointestinal diseases.
Uses
Pharmaceutical Industry (Main Use): Primarily used for the treatment of active duodenal ulcers, gastric ulcers, gastroesophageal reflux disease (GERD), radiation proctitis, and gastric inflammation; it can also prevent stress ulcers. It exerts a therapeutic effect by forming a protective film on the surface of damaged gastric mucosa, resisting the invasion of gastric acid and pepsin, adsorbing pepsin to inhibit its proteolytic activity, and promoting ulcer healing.
Clinical Auxiliary Application: Used to relieve stomach pain, heartburn, and acid reflux caused by excessive gastric acid in chronic gastritis; also applicable for the treatment of esophageal ulcers and gastritis induced by non-steroidal anti-inflammatory drugs. It is available in various dosage forms such as tablets, capsules, suspensions, chewable tablets, and granules, which are convenient for different groups of patients to take.
Research Application: In vitro studies have shown that it can protect intestinal epithelial cells from damage caused by acid and pepsin, and may interfere with the adhesion of intestinal epithelial cells to collagen I, which is under further research for related clinical expansion applications.
Molecular Formula & Structure
- Molecular Formula: C??H??Al??O??S? (basic formula); or Al?(OH)??(C??H??O??S?)(Al(OH)?)?(H?O)? (x=8~10, y=22~31, hydrous form)
- Molecular Weight: Approximately 2086.7 g/mol (basic form); 2483–2801 g/mol (hydrous form)
- IUPAC Name: Hexadeca-μ-hydroxy tetracosa hydroxy(μ?-(1,3,4,6-tetra-O-sulfo-β-D-fructofuranosyl-α-D-glucopyranoside tetrakis(hydrogen sulfato)?-))hexadecaaluminum(2)
- Synonyms: Sucrose octasulfate aluminum salt; Ulcerlmin; Sulcrate
Physical & Chemical Properties
- Appearance: White to slightly yellowish white amorphous powder; odorless, tasteless, with strong hygroscopicity; no visible impurities.
- Melting Point: >220 °C (decomposition, lit).
- Boiling Point: 1990 °C at 101.325 kPa (atmospheric pressure).
- Density: Approximately 0.338 g/cm3 (at 20 °C).
- Solubility: Almost insoluble in water, ethanol, and chloroform; soluble in dilute hydrochloric acid and sodium hydroxide solutions; slightly soluble in dilute sulfuric acid.
- Stability: Stable under normal temperature and dry conditions; sensitive to moisture and high temperature; decomposes when heated above 220 °C; stable in acidic environment, easy to dissociate and polymerize.
- Chemical Traits: It is a basic aluminum salt of sucrose octasulfate, with a complex structure containing aluminum ions and sulfate groups; it can dissociate into negatively charged sucrose octasulfate complex ions in acidic environment, which polymerize into insoluble colloids.
- Toxicity: Low acute toxicity; common adverse reaction is constipation; long-term and high-dose use may lead to phosphorus loss, causing hypophosphatemia and osteomalacia. It is contraindicated in patients allergic to sucralfate, premature infants, and immature neonates; caution is required in patients with habitual constipation, liver and kidney insufficiency, or dialysis patients; lactating women should not take it.
Synthesis & Storage
- Synthesis: Prepared by the reaction of sucrose octasulfate with aluminum hydroxide under controlled conditions, followed by purification, drying, and crushing processes; the product quality meets USP and CP standards, suitable for industrial production of pharmaceutical preparations.
- Storage: Sealed in dry, airtight containers; stored at -20 °C in a dark, dry place to avoid moisture, high temperature, and direct sunlight; shelf life is about 2 years when stored properly; avoid contact with strong acids and strong bases.
- Safety: Operate in a well-ventilated and dry environment; wear sterile protective gear (gloves, goggles, mask); avoid inhalation and skin/eye contact; wash hands thoroughly after handling; if accidentally inhaled or in contact with eyes, rinse immediately with plenty of water and seek medical attention if irritation persists; dispose of waste in accordance with pharmaceutical waste regulations.
