Product Description
Bupivacaine hydrochloride (CAS: 14252-80-3) is a high-purity amide-type long-acting local anesthetic, appearing as a white to off-white slightly hygroscopic crystalline powder (odorless, tasteless). Related to lidocaine and mepivacaine, it has strong potency and long duration, with minimal effects on respiration and circulation at recommended doses. Synthesized via alkylation and salification, it is widely used in clinical anesthesia, postoperative analgesia, and pharmaceutical/laboratory research.
Uses
Clinical Anesthesia: Used for local infiltration, peripheral nerve block, caudal and lumbar epidural block (surgery, dentistry, obstetrics). Only 0.25% and 0.5% concentrations for obstetrics; not recommended for intrathecal or intravenous regional anesthesia.
Postoperative Analgesia: Continuous epidural analgesia to relieve postoperative pain, reduce opioid use, and improve recovery comfort.
Pharmaceutical Research: Standard reference substance/intermediate for local anesthetic R&D, optimizing formulations and studying anesthetic mechanisms.
Laboratory Research: Used in biochemical/pharmacological experiments (nerve conduction, tissue metabolism); laboratory use only, not for clinical/household use.
Molecular Formula & Structure
- Molecular Formula: C??H??N?O·HCl (anhydrous); C??H??N?O·HCl·H?O (monohydrate)
- Molecular Weight: 324.89 g/mol (anhydrous); 342.91 g/mol (monohydrate)
- IUPAC Name: 1-Butyl-N-(2,6-dimethylphenyl)piperidine-2-carboxamide hydrochloride
- Synonyms: Marcaine; Bupivacaine HCl; MFCD00012100; EC 627-065-3; RTECS TK6125000; UN 2811; Customs Code: 2933399090.
- CAS Number: 14252-80-3
- SMILES Code: C1(CCCCN1CCCC)C(=O)NC1C(C)=CC=CC=1C.Cl
- Chemical Structure: Amide-type anesthetic with a piperidine core, butyl group at 1-position, carboxamide at 2-position, and 2,6-dimethylphenyl on amide nitrogen. Hydrochloride form improves water solubility; blocks sodium ion channels to inhibit nerve conduction.
Physical & Chemical Properties
- Appearance: White to off-white crystalline powder, odorless, tasteless, slightly hygroscopic; injection is clear colorless solution (discard if discolored/particulate).
- Melting Point: 249–251°C (anhydrous); 255–256°C (lit.); pKa: ~8.12; non-flammable solid (boiling point/density/flash point unreported).
- Solubility: Freely soluble in water and 95% ethanol; slightly soluble in chloroform/acetone; poorly soluble in phosphate buffers. Aqueous solutions without epinephrine are autoclavable; those with epinephrine require light protection and cannot be autoclaved.
- Stability: Stable under normal temperature and pressure; slightly hygroscopic, avoid moisture and prolonged light exposure.
Synthesis & Storage
- Synthesis: Mainly via alkylation of 2,6-dimethylaniline and piperidine derivatives, followed by salification with hydrochloric acid; crude product purified by recrystallization to reach pharmaceutical grade.
- Storage: Store at 15–25°C in a cool, dry, well-ventilated warehouse; keep sealed to prevent moisture absorption. Packed in sealed glass bottles or plastic drums. Avoid light, high temperature, and contact with oxidants; keep away from food/feed. Shelf life: 24 months (unopened, proper storage).
- Handling Precautions: Use in well-ventilated areas; wear protective equipment to avoid skin/eye contact and inhalation. In case of contact, rinse thoroughly with water; seek medical attention if irritation persists. Dispose of waste per local regulations.
